Have purchased a used ruger single 6 with only mag cyl, Can 22 long rifle be fired , if no , why not. you can shoot 22 short and 22 long rifle, in most 22 long rifle chambered rimfires?

No. You need the 22 lr cyl. I have the same issue in reverse. I lost my Mag cyl in a move years ago.

My Ruger Single Six only came with the 22lr cylinder.Just check out all the common gun auction sites and click on to the gun parts section then use the key words"Ruger Cylinder" and pick one out.It worked for me and dropped right in.:)

Try putting a LR shell in that cylinder.

This. 22LR has smaller diameter than 22WMR. It will go in, and it will fire, but you're asking for case failure.

The Lr will fire in that cylinder, but will swell and be harder to eject. The forcing cone will center the bullet ok where I didn't feel any splatter. Not a good practice tho. Much easier to find the LR cylinder than a mag cyl.
